Orrette Wauchope completed his B.Sc. degree in Chemistry at Brooklyn College of the City University of New York (CUNY). After graduating, he moved to Maryland to pursue his Ph.D. in organic chemistry at the University of Maryland-Baltimore County working with Katherine Seley-Radtke. His Ph.D. work focused on the design, methodological development and the synthesis of chemotherapeutic agents possessing anticancer, antiviral and antiparasitic activities. Following his doctoral work, he joined the laboratory of Lawrence Marnett at Vanderbilt University. At Vanderbilt, his work focused on understanding the chemistry and biology of the formation of DNA adducts that are derived from lipid and/or DNA peroxidation. These adducts can then be used as means of determining their roles in mutation and carcinogenesis. He joined the Baruch faculty as an assistant professor in the fall of 2017. At Baruch, Dr. Wauchope is interested in understanding the chemical biology of quorum sensing (cell-cell communication). Quorum sensing has been implicated in several deleterious processes that are hallmarks of microbial infections in the body. Consequently, work in his lab is focused on developing chemical agents that disrupt this communication mechanism as well as to investigate the processing of the chemical signals that enable cell-cell communication. He is also interested in understanding the relationship between bacterial communication and DNA damage. He also has several on-going research collaborations within the department as well as researchers at other universities.

This project involves the use of a novel chemical biology approach to identify the protein(s) involved in biofilm formation. This strategy couples alkynyl azido-biotin capture for the isolation of adducted protein(s) with a photochemical release of the adduct from streptavidin. This will allow for the identification of the protein target(s) and also give some information about the mechanism of action. This will be crucial for the development of more potent inhibitors. The work outlined in this proposal is at the intersection of chemistry and biology and will be conducted by undergraduate students.

This project involves the use of organic chemistry to understand the effects of oxidative stress on bacterial communication. Understanding the impact of DNA damage on bacterial communication would add to the sophistication of our approach to understanding this interesting phenomenon of bacterial communication. Also, this research has the possibility to offer more avenues of combating bacterial crosstalk.

This project involves the design and synthesis of a new class of inhibitors featuring a lactam and a exocyclic conjugated vinylic bromide. Two series of first generation compounds will be synthesized and their effects on biofilm formation and/or biofilm dispersal will be evaluated

Previous reports have indicated anecdotally that DNA damage plays a role in bacterial communication and biofilm formation. In our present study we wish to definitively investigate the realtionship between DNA damage and biofilm formation using chemical agents that cause specific types of DNA damage. The effects of these agents on biofilm formation will be quantified and also visualized using confocal microscopy.
